Working with the spectrophotometer.

Figure 1: Spectra of a diluted series of Synechosystis sp, PCC 6803 and Nannochloropsis CCMP 1779.

We took cells from agar plates and diluted them with water. The spectra was then obtained using a spectrophotometer (infinite M200PRO). The data was saved at the lab computer as SpecScan1.xlsx.

The cuvettes containing Synne6803 (1:10) and Nanno1779 (1:20) was put in a refridgerator for later color comparison, as the color they had showed a good absorption level for measurements.

Figure 2 shows that there is very little difference between plastic and quartz cuvettes, even in the UV-region. The absorption for Nanno 1779 is slightly shifted upwards in the plastic cuvettes, while there was no significant difference between the glass and quartz cuvette for the Synnecho 6803.
