determination of the dry weight

1. 47-mm glass fiber paper filters (Whatman GF/C, Schleicher & Schuell Co.) were presaturated with 100 ml distilled water and dried overnight at 60°C and 1h at 120°C, to evaporate all the water. Eppendorf tube containing a presaturated paper filter were pre- weighted. 2. Replicate aliquots (10 ml) of culture suspensions were centrifuged (4700xg, 10 min, room temperature). 3. washed with 80…

Changes in lipid composition induced by environmental and genetic manipulation

We will investigate whether oxygen depletion will change the lipid composition in Synechocystis sp. PCC 6803 using chromatography and mass spectrometry techniques. Several enzymes with homology to enzymes involved in ubiquinone biosynthesis in other organisms appear to be involved in plastoquinone synthesis in cyanobacteria. We will investigate the functions of open reading frames with homology to ubiquinone synthesis in other…

First platereading

Two centrifuge tubes (50 ml) where filled with media and algaecells (Nannochloropsis CCMP 1779) that had been growing for 3 days. The tubes where then centrifuged at 6000 rpm for 5 minutes, and some of the supernatant was taken away. This was done to concentrate the cells approximately 8-folds. The solution was then transferred to a 24-well plate to make…

Working with the spectrophotometer.

We took cells from agar plates and diluted them with water. The spectra was then obtained using a spectrophotometer (infinite M200PRO). The data was saved at the lab computer as SpecScan1.xlsx. The cuvettes containing Synne6803 (1:10) and Nanno1779 (1:20) was put in a refridgerator for later color comparison, as the color they had showed a good absorption level for measurements.…
